[Association between TP53 codon72 and smoking and somatic mutation: Focusing on somatic mutation and TP53 codon72 in patients with bladder cancer, the frequency of FGFR3 was significantly higher in patients with non-muscle invasive cancer and Arg / Arg and Arg / Pro. It was We also found that the frequency of PIK3CA was significantly higher in Arg / Arg patients and the frequency of RAS mutations was significantly higher in patients with Pro / Pro alleles. In muscle-invading cancers, the mutation frequency of FGFR3 tended to be high in Arg / Arg and Arg / Pro patients as in non-muscle-invading cancers. On the other hand, no correlation was found between the mutation frequency of RAS, PIK3CA and TP53 and TP53codo72. Comparing the correlation between smoking and somatic mutation, smokers tended to have a high frequency of RAS mutations in both non-muscle invasive and muscular invasive cancer patients. When smoking and TP53 codon 72 allele information were integrated and compared, in non-muscle invasive cancer, FGFR3 mutations were high in patients with Arg / Arg and Arg / Pro alleles of TP53 codon 72, but RAS mutations were mostly found in nonsmokers. TP53 codon72および喫煙と体細胞変異との関連:膀胱癌患者の体細胞変異とTP53 codon72に着目し、筋層非浸潤癌においてはArg/ArgおよびArg/Pro患者においてはFGFR3の頻度が有意に高かった。また、Arg/Arg患者ではPIK3CAの頻度が有意に高く、Pro/Proアレルを持つ患者ではRAS変異の頻度が有意に高いことを見出した。筋層浸潤癌においては筋層非浸潤癌と同様でArg/ArgおよびArg/Pro患者においてはFGFR3の変異頻度が高い傾向を認めた。一方RASやPIK3CAおよびTP53の変異頻度とTP53codo72との間には相関を認めなかった。喫煙と体細胞変異との相関を比較すると喫煙者においては筋層非浸潤癌患者および筋層浸潤癌患者双方においてRAS変異の頻度が高い傾向を示した。喫煙とTP53 codon72のアレル情報を統合して比較検討すると筋層非浸潤癌においてはTP53codon72のArg/ArgおよびArg/Proアレルを持つ患者ではFGFR3変異が多いが、非喫煙者ではRAS変異がほとんど見られなかった。Pro/Proアレルを持つ患者では喫煙歴にかかわらずRAS変異が多い結果であった。ジャポニカアレイによる網羅的SNP解析:ジャポニカアレイを用いて網羅的SNP解析を96例について解析を終了している。体細胞変異陽性患者および陰性患者を比較して、変異の頻度に関連するSNPの検索を進めている。
